    Why is deep sleep such a big deal, as long as you get around 7 hours total sleep a night, shouldn’t that be enough?

    Deep sleep has an ancient, restorative power to clear waste from the brain, it’s our body’s way of taking out the trash; waste clearance occurs whilst we are awake but is substantially enhanced during deep sleep.

    This waste potentially includes toxic proteins that may lead to neuro-degenerative diseases such as Alzheimer's disease and Parkinson's.

     

    My Sleep Statistics 2019

    You can see here that my sleep on 29th, 30th and 31st of October 2019 was good as far as overall time is concerned, 7:34, 7:49 and 7:10.

    But my deep sleep; the most rejuvenating sleep only accounted for 8% and 17% of the overall total.

    I managed to increase that to over 25% and if some cases over 40%, and now I will tell you how.

     

    Fitness Tracker

    As part of my NMN Experiment I purchased a fitness tracker to monitor my heart rate, another function of this tracker is a sleep monitor, it records overall sleep, light sleep, deep sleep and minutes awake.

    Link below to the one I use...I may do a sperate video on it, let me know if you’d like that.

    I Experimented with different sleep aids, techniques, types of food, when I ate food, alcohol intake etc.

    Unless there is an unexpected occurrence, I can now pretty much guarantee that I will get 7 to 8 hours of sleep a night, but more importantly that over 20%, (in one case 43%) will be deep, rejuvenating sleep.

     

    SMART Lights

    With white, blue and green light being such an issue to our alertness and melatonin levels I stared to research smart light bulbs, bulbs that I could programme to dim and gradually turn red 2 hours before I actually went to bed.

    When the lights are on after 8pm it reminds me of the Ops Rooms that I would often frequent when I was in the Army…no white light after sundown.

    I used these in my study, the living room and the bedroom, in conjunction with my blue light blocking glasses (which I will cover later).

    Blue and Green Light Blocking Glasses

    I listened to the likes of Dave Asprey who extoled the virtues of Blue and green light blocking eyewear, of which he owns a company,

    I also watched an interview with the BLUblox CEO, Andy Mant where he further explained the science behind how blue light affects our sleep patterns.

    The science seemed unquestionable, so I purchased this pair of glasses.

    Did they work, well when you put them on 2 hours before you sleep, they certainly do make you feel sleepy.

    However, over a period of a few months I tried wearing them on and off and made sure that all lights were out before I went to bed and only had the red lights on if I had to go to the toilet during the night.

    They made absolutely no difference to my overall sleep, but more importantly to my deep sleep. This may be different for you, Dave Asprey or Andy Mant, but for me ZERO effect!

    So, they are now in the cupboard of broken dreams.


    Eye Mask

    The next thing I tried was an eye mask; I bought this one from Amazon.

    Again I tried this with

    ·     The SMART lights and nothing else

    ·     The blue light blocking glasses and nothing else

    ·     The smart lights and the glasses

    ·     And with just the eye mask and bright white light until I got into bed

    This did improve my deep sleep considerably and it allowed me to walk around the house, watch TV and use my smart phone without having to wear the blue light blocking glasses.

    So, this $13 eye mask is now a consistent part of my nighty routine.


    Blackout Curtains

    I had blackout curtains in our bedroom, but there was still a certain amount of light that bled in, this is because there were 2 very large floor to ceiling windows in the bedroom.

    When we moved to a new house that had 1 smaller window that was completely covered by the curtains, again my deep sleep improved.

    I think that the Blackout curtains and blackout eye-mask are the biggest contributors to my extended deep sleep.

     

    Sleep Music

    Listening to sleep music or sleep noises certainly helped when going to sleep an there are many studies that bear this out. I found that I fell asleep quicker when I had music on in the background, and the most relaxing was actually thunder and rain, I now have 4 that I like to rotate through.

    I will upload the black screen videos I found most soothing so you can try them out too.


    Room Temperature

    It took me about 2 months to get a temperature I liked, the a/c is now set to 23 degrees C, I use this to check it every night.

    Dr. Matthew Walker states it is a scientific fact that your body and brain need to drop their core temp by around1°C (2 to 3°F) to initiate sleep, that’s why it is easier to fall asleep in a room that is too cold and not too hot.

    I also take a warm shower before I go to sleep, this ensures that my blood rushes to the extremities and away from my core, when I get out of the shower there is then a massive dissipation of heat, out through the extremities and my core temperature plummets, so this adds to the cooling effect that everybody needs to sleep.

     

    Food and Drink

    Again, this took many months of adjusting the time I would eat and drink before going to bed. Also, what I would eat, was it a snack or a full meal, was a fat heavy meal or Carb heavy meal.

    This was the most difficult; one night for example I would eat steak at 7.30 pm and have 25% of my sleep that night be deep; I would replicate it the next night only to find my deep sleep went down to 18%.

    It wasn’t until I had perfected the dark sleep environment (curtains and mask), the relaxing music and the temperature that I realised that, what I ate and when I ate it made very little difference to my deep sleep levels at all.

    Even alcohol, on the weekend I could drink 1 or 2 glasses of wine right up to 10 pm and still get over 25% deep sleep and no reduction in overall sleep either.

    I think 1 or 2 glasses of wine, once or twice a week, over the period of a few hours isn’t excessive, that said, on special occasions like birthdays I have drunk more, and it has affected my overall and deep sleep.

    Remember my sleep on 29, 30 and 31 of October 2019 was good as far as overall time is concerned, 7:34, 7:49 and 7:10, but my deep sleep only accounted for 8% and 17% of the overall total.

    Here on the 3rd, 4th and 5th of August 2020, even on two nights when I only had just over 5:30 minutes total sleep, my deep sleep was between 28% and 34%, up from 8% and 17%.

    This marked change took me about 10 months to achieve.

    And just to show that it is still working, my sleep Stats from the 19th, 20th and 21st of January 2021 are (I am preparing and recording this video on the 21st of Jan).

    My overall sleep was 7:36, 6:57 and 8:07

    With deep sleep constituting 29%, 36% and 34% respectively  

     

    Summary

    So, if you too want to improve, not only your overall sleep, but far more importantly your deep, regenerating sleep, I would try these things in this order (show pictures and talk):

    For adults in their 40s, sarcopenia is hardly noticeable—about 3% muscle mass is lost each decade. However, for those aged 65 years and older, muscle decline can become much more rapid, with an average loss of 1% muscle mass per year.

    More importantly, sarcopenia is also marked by a decrease in strength, impaired gait, reduced physical activity and difficulty completing simple everyday tasks such walking, sitting down and standing up unaided.


    The Future

    The proportion of older adults aged 65+ is projected to more than double by the year 2060, this is driving research into the process of musculoskeletal decline.

    Researchers at Colorado State University's Columbine Health Systems Center for Healthy Aging believe they have found an animal model that will help them better understand it and find ways to limit the symptoms.

    The study, published in Frontiers in Physiology: Striated Muscle in October, is an example of using comparative medicine to understand human diseases and conditions.

       

    Animal Models

    Scientists often rely on animal models to mimic disease progression and study the prevention, diagnosis and treatment of medical conditions.

    However, until now, no animal model has been able to fully capture all aspects of human musculoskeletal aging.


    Karyn Hamilton

    Karyn Hamilton, a professor in the Colorado State University's Department of Health and Exercise Science, and associate director at the Center for Healthy Aging said "Existing pre-clinical models either rely on unloading the muscle, meaning mimicking bedrest in an animal, or they must wait until the animals get really, really old, and even then they don't really get the same muscle aging phenotype as people do."


    Dunkin Hartley Guinea Pig

    In their work, Hamiliton's team found that the Dunkin Hartley guinea pig was a good candidate for a muscle aging model due to the animal's tendency to develop osteoarthritis at a young age.

    The two conditions, osteoarthritis and sarcopenia, seem to be linked in humans; with advancing age, skeletal muscle dysfunction increases the risk for osteoarthritis, and osteoarthritis increases the risk for further muscle decline.


    Hamilton’s team

    Hamilton teamed up with Dr. Kelly Santangelo—an associate professor in the Department of Microbiology, Immunology, and Pathology at CSU, who has been studying primary osteoarthritis in Hartley guinea pigs for many years, also Associate Professor Raoul Reiser, also in Department of Health and Exercise Science, to understand how skeletal muscle changes as osteoarthritis progresses in guinea pigs.

    The team hypothesized that those muscle changes may mimic human musculoskeletal aging.

     

    Classic signs

    Muscle composition also changes with age: fast-twitch fibers, which are larger and capable of exerting stronger forces, decrease in number, and slow-twitch fibers, which are smaller and less metabolically efficient, tend to increase.

    Generally, when people "lose muscle mass," it's the large, fast-twitch fibers that are affected, also with decreasing muscle mass tends to come an increase in fat mass.

     As humans age, it may be that maintaining muscle function is more important than preventing loss of mass, so does this mean weight training and functional exercise as opposed to just bulking up with muscle? As an aside, I know many body builders who have plenty of muscle, but have a pondered gait and can’t scratch behind their neck.


    Statement by Hamilton

    "What we've learned over the decades is that age-related loss of muscle mass and age-related loss of muscle function don't always go hand in hand, you can do things to improve muscle mass, and you might not get an improvement in muscle function; you can do things to improve muscle function and may not get an increase in muscle mass."

     

    Comparisons

    The researchers compared muscle changes observed in Hartley guinea pigs with those in Strain 13 guinea pigs; Strain 13 guinea pigs tend to develop osteo-arthritis later in their lifespan and, therefore, might not exhibit the same classic signs of muscle aging.

    They found some striking similarities to human muscle aging, such as a decrease in muscle density, likely due to an increase in fat mass.

    While a decrease in muscle mass was not noted, researchers did find a shift toward smaller, slow-twitch muscle fibers, as is expected in human muscle with advancing age.


    Hamilton

    "If you look at the overall picture, we think that some of the key things that always happen with human muscle aging—that shift toward a less powerful, slower-twitch muscle phenotype—are quite clearly modeled in the Hartley guinea pigs. And we believe that if we started looking at even older guinea pigs, we might see more of the things that people think of as classic sarcopenia."

     

    Future Directions

    This study now provides a baseline that allows the team to take multiple directions in future research.

    One direction will be to employ functional tests to study how muscle strength and gait, or mobility change with age in the Hartley guinea pigs, and how these changes mimic the deterioration of muscle function in aging humans.

     A priority will be to identify if Hartley guinea pigs can be a valuable translational model for identifying interventions that show promise for preventing or slowing the decline in overall musculoskeletal function in aging in humans.

    At the 249th National Meeting & Exposition of the American Chemical Society (ACS) it was announced that supplementing with Vitamin D supplements may slow or even reverse the progression of less aggressive, or low-grade, prostate tumors without the need for surgery or radiation.

       

    Gleason Grading System

    If a tumor is present in a prostate biopsy, the pathologist will grade its aggressiveness on a scale known as the Gleason Grading System.

    Tumors attracting a score of 7 or above are considered aggressive and are prone to spreading; they will require either surgical removal of the prostate gland or radiation therapy.

    However, tumors with a score of 6 or less are classed as less aggressive and in some cases may show no symptoms or cause no health problems for the duration of the man's life.

    Active Surveillance

    Bruce Hollis, Ph.D., who is at the Medical University of South Carolina, explained that in cases of a low-grade prostate cancer (a score of 6 or below), many urologists do not treat the disease, but instead initiate "active surveillance. 

    The cure, which means surgery or radiation, is probably worse than the disease, so urologists will wait a year and then conduct another biopsy to see where the patient stands."

    Elective Surgery

    That said, knowing that they have even low-grade prostate cancer can cause patients and their family’s excessive anxiety, this may prompt some of the men to undergo elective surgery, despite the risk of complications such as infection, urinary incontinence and erectile dysfunction.

    But a man must wait at least 60 days from the time of his biopsy before he can undergo surgery, this is that any inflammation caused by the biopsy can wane.


    Vitamin D supplementation

    Bruce Hollis wondered if giving these men vitamin D supplements during the 60-day waiting period would affect their prostate cancer.

    Previous research had shown that when men with low-grade prostate cancer took vitamin D supplements for a year, 55% of them showed decreased Gleason scores, and in some of the cases the tumors disappeared completely.

     

    Clinical Trial

    In a new randomized, controlled clinical trial, Bruce Hollis’s team was assigned 37 men who were due to undergo elective surgery.

    There were two groups, one that received 4,000 IU of vitamin D per day and a second placebo group that didn't receive vitamin D.

    The men's prostate glands were removed and examined 60 days later.

    Results

    Hollis stated, “The new research suggests that vitamin D supplementation may improve low-grade prostate cancers by reducing inflammation, perhaps lessening the need for eventual surgery or radiation treatment.”

    He went on to say…

    ·"We don't know yet whether vitamin D treats or prevents prostate cancer, at the minimum, what it may do is keep lower-grade prostate cancers from going ballistic."

    This is a review of the article ‘Another Win for Senolytics: Fighting Aging at the Cellular Level Just Got Easier’ by Shelly Fan PhD and some other interesting information on senolytic trials.

    Links in the description below to the article and the other links I used to make this presentation.

     

    Hallmarks of Aging

    In the article Shelly references the parable of the blind men and the elephant and likens it to Longevity. For those unfamiliar with the parable, in essence blind men who don’t know what an elephant is are asked to touch a different body part and then conceptualize what the animal is like, because of their limited experience and specific area of touch, each man has vastly different conclusion and they all believe that they’re description is correct. She says that aging, thanks to its complexity, is the bio-medical equivalent of the elephant.

    In that researchers have focused on either one or another “hallmark” of aging, with some success, but not a number together.


    Joining the Dots

    For example, past research has shown us that energy production in our cells can become erratic and non-functional as we age. Cells programmed for apoptosis don’t die, but instead become zombie-like “senescent cells.

    A "senescent cell" is a cell in our body whose telomeres have reached their "Hayflick Limit" i.e. its telomeres have become so short that the cell cannot divide anymore.

    It is not a healthy cell anymore, it’s not exactly dead, but it is programmed for death, this cell death is called apoptosis, the Greek for self-eating, if it doesn’t die it starts to produce chemicals that increase inflammation and damage our DNA.

    The key is to find out how other hallmarks of aging, such as lower NAD, inflammation and shortening telomeres interact with zombie cells, so we can see the whole picture.

     

    The Study

    A new study published in Nature Metabolism has starting to connect the dots. A study, in mice, linked-up three promising anti-aging pathways

    ·     Attacking senescent cells

    ·     Addressing inflammation

    ·     Tackling unreliable energy production in cells

    All three elements point to one key factor that drives aging.

     

    Healthy to Zombie Cell

    Shelly likens individual cells to small cities; each with their own power plants and their ‘superstar’ molecular worker is Nicotinamide Adenine Dinucleotide (NAD). As we know NAD is a molecule that’s critical for helping mitochondria produce energy.

     As we age our cells start losing NAD and with reduction our mitochondria become less able to produce the energy we need, which in turn flips normal cellular function into dysfunction. At present this hypothesis has only been prove in mice.

    One dysfunctional element is that cells programmed to die through apoptosis don’t die, are not consumed by the body as extra fuel, but instead hang-around as senescent or zombie cells.

    These zombie cells then turn to the dark side, and start to leak an inflammatory cesspool of molecules called Senescence Associated Secretory Phenotypes (SASP) that “spread” harm and injury to adjacent healthy cells.

      

    Senolytics

    So, let’s look at senolytics. Senolytics are a group of compounds that destroy these “zombie” cells.

    A study in old mice, the equivalent of a 90-year-old human, found that wiping out these zombie cells with two simple compounds increased their lifespan by nearly 40 percent.

    Others using a genetic “kill switch” in mice found that destroying just half of their zombie cells helped the mice live 20% longer and also allowed them to have healthier kidneys, stronger hearts, more luscious fur, and higher than normal energy levels.

    In the same way that some companies now sell NAD boosters like NMN and NR, pharmaceutical companies are investigating over a dozen potential senolytics in a race to bring one to market.

    But is two better than one, could NAD boosters and senolytics be combined?


    The Buck Institute

    This new study, led by Dr. Judith Campisi and Dr. Eric Verdin at the Buck Institute for Research on Aging in California, asked if the lines between lower NAD and zombie cells could be connected.

     And asked could the connection be CD38?

    If you have watched my video on raising NAD and lowering CD38, you will know that CD38 wreaks havoc by both boosting inflammation and destroying our beloved NAD.

     

    CD38 Study

    Using tissue from mice and humans, the team traced CD38 to a type of immune cell. These cells, called M1 macrophages (which literally means, ‘big eaters) are well known to increase inflammation in the body and cause DNA damage as we age.

    When comparing fat tissue isolated from young and old mice, the team realized that M1 macrophages relentlessly pump out CD38 as the cells age, which in turn, breaks down the much-needed NAD.

    Dr. Eric Verdin used the analogy of a sink and water source and said "Do NAD levels drop because of a faucet problem, i.e. our ability to make NAD, or is it a leaky sink problem, where aging cells break down NAD too fast. Our data suggests that, at least in some cases, the issue stems from the leaky sink.”

    Which to me means we can make NAD okay; it’s just that it runs out before we can use it.


    The Zombie Connection

    This is what we know so far: aging triggers immune cells that pump out CD38, a chemical that destroys our NAD.  But, how can we stop CD38?

    In an unexpected twist of events, the connection actually seems to be the zombie cells.

    Remember, zombie cells leave a chemical evidence trace called Senescence Associated Secretory Phenotypes (SASP). They also change their ‘molecular structure’ so it’s possible to separate them from the healthy cells.

     In the fatty tissue gathered from aged mice, the team identified zombie cells and found that their “toxic waste” vastly increased the amount of CD38.

    So, it is the Senescence Associated Secretory Phenotypes (SASP) that direct the immune cells to make more CD38.

     

    Testing the Theory

    So, if zombie cells are the directors, then getting rid of them should reduce CD38 and in turn, preserve our NAD; well that’s the theory.

    To test it out, the team used genetically engineered mice; this allowed the scientists to identify the zombie cells and then selectively kill them.

    The team injected the mice with a drug that damaged their DNA. This mimicked aging, in the sense that it increased zombie cells and CD38.

    Result, killing the zombie cells lowered CD38 levels, which in turn preserved NAD.


    Statement

    Dr. Verdin said “We are very excited to link two phenomena which have been separately associated with aging and age-related disease.

    For now, zombie cells seem to be a master-level culprit that drives inflammation, decreases NAD levels, and breaks the cell’s energy production.

    This suggests that senolytics, which selectively kills off zombie cells, could as a secondary effect also increase NAD, something we didn’t know previously."

     

    NAD Boosters

    On supplementation Dr. Eric Verdin said, “Ultimately I think supplementation will be part of the equation, but filling the sink without dealing with the leak will be insufficient to address the problem.”

     

    In other words, for NAD supplementation to work more efficiently, we may need to also use senolytics drugs to decrease zombie cells and CD38 levels; in essence plugging the leak in the sink, before we turn the NAD tap on


    Where to get Senolytics?

    In an article posted in the US National Library of Medicine, it states that Senolytics are a class of drugs that selectively clear senescent cells.

    ·     The first senolytic drugs

    o  Dasatinib (approved in the USA and EU as a leukemia treatment)

    o  Quercetin (a plant Flavonol available in supplement form)

    o  Navitoclax (an experimental anti-cancer drug)

    o  Fisetin (a dietary Flavonoid available in supplement form)

     

    Early pilot trials of senolytics suggest they decrease senescent cells, reduce inflammation and alleviate frailty in humans

    Clinical trials for diabetes, idiopathic pulmonary fibrosis, Alzheimer’s, COVID‐19, osteoarthritis, osteoporosis, eye diseases, bone marrow transplants and childhood cancer survivors are either underway or just beginning.

    Sarcopenia is the loss of muscle mass specifically related to aging. It’s normal to lose some muscle mass as you age. However, sarcopenia describes severe muscle loss that strays from the norm. Sarcopenia affects your gait, your balance and your overall ability to perform daily tasks. For too long, researchers have believed that this deterioration in muscle was inevitable. But they’re now beginning to look into treatments that might prevent or slow down this process.

     

    Symptoms

    People with sarcopenia often experience weakness and lose stamina. This can affect their ability to carry out even the simplest of physical activities like walking or sitting. This reduction in activity then leads to further muscle mass loss and instability.

     

    Causes

    A common cause of sarcopenia is decreased physical activity throughout the day. However, although less frequent, some people with active lifestyles may also be diagnosed with sarcopenia, this suggests that there could be other reasons for the development of the disease.

     Researchers currently believe that other causes of sarcopenia could include:

    ·     a reduction in the nerve cells that send signals from your brain to tell your muscles to move

    ·     a lowering of your hormone levels

    ·     a decline in your body’s ability to convert protein to energy, remember this is one of the roles of NAD

    ·     not consuming enough daily calories and protein in order to maintain your muscle mass

     

    Treatment - Exercise

    The main treatment path for sarcopenia is exercise. Researchers have identified resistance training as the specific form of exercise that is most beneficial to people with sarcopenia. This training is designed to improve muscle strength and stamina and uses resistance bands or weights.

     Resistance training can also help balance your hormone levels.

    It has been shown to improve our ability to turn protein into energy. These changes have in some cases been seen in only two weeks.

     The right intensity and frequency of exercise is vital, so you achieve the best results, without causing injury.

    So, spending 45 minutes at the gym, but only training for 25 won’t cut it; trust me you’re only kidding yourself .

     

    Hormone Replacement Therapy (HRT)

    HRT can help to raise lean body mass, decrease abdominal fat, and prevent bone loss in women whose hormone levels have decreased during the menopause. However, the use of HRT is debatable, mainly because of an increased risk of some cancers and other severe health conditions.


    Other Treatments

    Some other treatments that are under investigation include:

    ·     growth hormone supplements

    ·     testosterone supplements

    ·     Vitamin D, remember to supplement with Vitamin K2 also to ensure the calcium goes to your bones and teeth and not into your soft tissues

    ·     medications for the treatment of metabolic syndromes

    If these prove useful, they should be used in conjunction with resistance training, not in place of it.


    Prevention

    Lack of activity is the most common reason behind this condition. Just half an hour of moderate exercise each day, like jogging or even walking, will help keep your system working and fit. In order for exercise to be effective, proper nutrition is also important. 

    Research has shown that consuming more protein may help older adults reduce their chance of sarcopenia

    Supplements

    Supplements have also proven useful in the prevention of sarcopenia. Some include:

    ·      Creatine, for increasing and maintaining muscle mass (I take)

    ·      Vitamin D, for maintaining bone and muscle tissues (I take)

    ·      Protein, to help preserve body mass (don’t take)

    As before these should be used in addition to exercise and not instead of it.


    Associated conditions

    Sarcopenia has been linked to metabolic problems like:

    ·     type 2 diabetes

    ·     high blood pressure

    ·     Obesity

     These three conditions are a type of gateway, which put you at greater risk of the developing coronary heart disease, stroke and other conditions that affect the blood vessels.

    What is Epigenetics?

    Epigenetics is the science of how our DNA is controlled and how it’s affected by external factors; these external factors affect our health-span and lifespan.

    Genetics is fixed from birth; we get our genes from our parents and they define us as being male or female, they control our eye colour, height etc.

    This is what is measured by companies such as 23andMe.

    We all share more than 99% of our DNA, however the remaining 1% varies from person to person; this is what makes us unique

     Epigenetics looks into the dynamic changes in our gene function overtime, changes that don’t involve a change in the underlying DNA sequence; these changes are influenced by external factors

     

    Good to Know

    So, how much can these external ‘epigenetic’ factors actually affect out lifespan and health-span?

     Someone who is at high risk of heart disease from a genetic predisposition, so there is a family history of heart disease, may have four times the chance of developing heart disease because it is in their genes; that is obviously bad, but compare that to someone who has a low risk of heart disease from a genetic pre-disposition, but is affected by external environmental and lifestyle factors, the risk to them is 5,000 fold, so 5,000 times more chance of getting heart disease because of the environment or the life choices they made.

    So, there is a lot more risk from factors that are under our control, than from our genes,

     As David Sinclair says, ‘Only 20% of longevity is genetic, the rest is up to you, what will you do today?


    You can Affect this

    So, identical twins have the same DNA, but through lifestyle choices may well have vastly different health outcomes by the time they reach middle-age (Source 3), which twin smoked and didn’t use sunscreen?

    Biological age testing allows us to read our epigenetic markers, markers that we can influence through changes to our lifestyle.

     Epigenetic markers change every time one of our cells divides, and even when our cells are not dividing, changes are still taking place to our epigenetic markers.

     We can affect, through lifestyle, the way that these markers change and through DNA methylation testing we can monitor our progress, either good or bad.

     It is not easy to do the right thing and there is no magic pill, targeted supplementation is only a small part of the answer.


    Our Biological Age?

    So, why should we check our biological age? Our Epigenetic age is a much better measure of our health, well-being, lifespan and health-span than our chronological age.

    ​Knowing our epigenetic or biological age allows ‘us’ - you and me - to move our healthcare needs from being reactive to being proactive;

    We can become citizen scientists; we can take responsibility for our own longevity by measuring our health objectively at the molecular level

     And once we know our Epigenetic age, we can take measures to adjust it if necessary, I am guessing no matter what your biological age is, you would like to see it lower?

     

     The Slippery Slope

    Epigenetics measures how well your body is aging, if your epigenetic age is higher than your biological age you are more at risk from age related diseases and all-cause mortality [Source 2]

    David Sinclair often talks about gateway issues such as being pre-diabetic or having elevated blood pressure, these very rarely stand-alone forever, they usually open the door to type 2 diabetes and hypertension, which in turn lowers our bodies defenses and opens us up to even more serious diseases.

     A forty-year-old whose epigenetic age is fifty-five is considerably more at risk from age related diseases than say a forty year old whose epigenetic age is forty-two, this stands to reason.


    DNA Methylation v Telomere Lengths

    There are now many companies that measure telomere lengths to predict epigenetic age, this is Dr Tom Stubbs, the CEO of Chronomics a company that conducts DNA methylation tests.

    There are some issues with the accuracy of telomere length testing, but I will let Dr Tom Stubbs the CEO of Chronomics explain the details.

    So, there are upwards of 20 million markers available to build models that can more accurately predict age, so there is a lot more information available than the one marker that is used when estimating age using telomeres, this means that epigenetic testing is much more accurate for estimating someone’s biological age, so, in this case, bigger is better


    ​​Chronomics

    We have already heard from the CEO of Chronomics, Dr. Tom Stubbs, but in outline Chronomics are a B2B company who sell their technology to other companies; DoNotAge.org is one of those companies, they also have a YouTube channel where you can gather more epigenetic age testing information.

    You can see from this image that the number of biological markers used by Chronomics will give a significantly more accurate epigenetic age than that of their competition, this information was gathered by DoNotAge.org as they searched for the best B2B provider of an epigenetic age prediction service.

    The closest competitors are Muhdo with 850,000 markers and Elysium with 100,000 markers.

    The company at the bottom of the list, epi-age,com is the company that I used on my 56 birthday, before I knew what I know now, but mainly out of necessity, they were at the time the only company I could find that did international testing.

    Out of the 28 million available markers that can be measured to predict age, they use 13, not 13,000, just 13, so I am not too sure how many months margin of error that allows for, Chronomics with 20 million markers claim an accuracy bracket of within 6 months.

     Obviously, this level of science and accuracy comes at a cost, prices should come down with time, but at present the most accurate test is still very expensive

    A long-term clinical study from Denmark has shown that increased Nicotinamide Adenine Dinucleotide (NAD) levels and improved mitochondrial function results from NR supplementation in mice could not unfortunately be repeated in humans.

    NR 12 Week Study in Humans

    Studies in animals have shown that NAD boosters stimulate metabolic health by improving mitochondrial function in skeletal muscle, so far NR supplementation has only been shown to raise human NAD levels in blood.

    Scientists from Denmark have published, in the Journal of Physiology, the results of a 12-week study where they tested the effects of NR on NAD levels and mitochondrial function in human skeletal muscle.

     

    Metabolic Health

    Our metabolic health depends on mitochondrial function, but as our NAD decreases with age so does our mitochondrial function.

    Research has linked these declines with age-related diseases such as obesity, insulin resistance and type 2 diabetes, this in turn results in the defective regulation of our skeletal muscle metabolism.

     

    Scientific Study

    The team of scientists studied NR supplementation to see if it did in fact increase NAD levels and improve mitochondrial function in human skeletal muscle too.

    The clinical trial consisted of 40 participants (men with obesity and insulin resistance)

     They were given 1000 mg NR or a placebo twice a day for 12 weeks, 2000 mg per day is a lot, the advised dose is around 300 mg a day.

     

    Results

    The results showed that NR supplements did not improve NAD levels in the participants’ skeletal muscle.

    The study also concluded that NR supplementation did not affect the level of mitochondria in their skeletal muscle.

    The team also measured the levels of citrate synthase, an enzyme that correlates to mitochondrial levels, and unfortunately also found no changes.

    The team also checked to see if NR supplementation increased reactions that use oxygen to convert nutrients into cellular energy, they found that NR supplementation did not increase in the overall mitochondrial oxidative capacity in human skeletal muscle


    Study Conclusion

    The team stated, ‘our data does not support the hypothesis that dietary NR supplementation has significant impact on skeletal muscle mitochondria in obese and insulin-resistant men.’

    They also stated, ‘Future studies on the effects of NR on human skeletal muscle may include both sexes and potentially provide comparisons between younger and older people.’

      

    The NR community

    The NR community has been very quiet about this study? Charles Brenner who is usually very vocal (in his tweets) when it comes to the lack of NMN human studies or rubbishes anything linked to David Sinclair such as resveratrol, seems to have lost the ability to Tweet.

       

    What the study doesn’t tell us

    This study is very specific, if you are a man with obesity and insulin resistance, NR will not improve mitochondrial function in your skeletal muscle, IMHO anyone commenting on this study and not talking about the specifics of this study is just guessing.

     This study was conducted specifically in men with obesity and insulin resistance, so does NR work on skeletal muscle if:

    You are a woman with obesity and insulin resistance

    If you are obese, but not insulin resistant or vice versa

    If you are neither obese nor insulin resistant

     There still seems to be more questions than answers and let’s hope that in the future that these type of studies are also conducted with NMN

    and that they test people with other age related diseases, not just obesity and insulin resistance.

    As we know Vitamin D is extremely important for good health.

    It plays several roles in keeping your body’s cells healthy and functioning the way they should.

    Most people don’t get enough vitamin D, so supplements are common.

    However, it’s also possible — although rare — for this vitamin to build up and reach toxic levels in your body.

    Let’s look at 6 potential side effects of getting excessive amounts of this important vitamin.

     

    Deficiency

    Vitamin D is involved in calcium absorption, immune function, protecting our bones, muscles and heart health.

    It occurs naturally in food and is also be produced by your body when your skin is exposed to sunlight.

    Aside from fatty fish, there are few foods rich in vitamin D, so many foods have to fortified with Vitamin D.

    Very few people get enough sun exposure to produce adequate levels of vitamin D.

    Deficiency is very common, it's estimated that about 1 billion people worldwide don't get enough vitamin D. Get a blood test and only supplement with what you need.


    Toxicity

    However, taking extremely high doses of vitamin D3 for long periods MAY and I stress MAY lead to an excessive build-up in your body.

    Vitamin D toxicity occurs when blood levels rise above 150 nanograms per milliliter (ng/ml).

    Thankfully Vitamin D toxicity isn't common and occurs almost exclusively in people who take long-term, high-dose supplements without monitoring their blood levels. Get a blood test and only supplement with what you need.

     

    Elevated blood levels

    Achieving adequate levels of vitamin D in your blood may help boost your immunity and protect you from diseases like osteoporosis, cancer and in 2 recent studies against COVID-19 too; however, there isn't agreement on an optimal range for adequate levels.

    Although a vitamin D level of 30 ng/ml is typically considered adequate, the Vitamin D Council recommends maintaining levels of 40–80 ng/ml and states that anything over 100 ng/ml may be harmful.

     While an increasing number of people are supplementing with vitamin D, it's rare to find someone with very high blood levels of this vitamin.

     

    Medical studies

    One recent study looked at data from more than 20,000 people over a 10-year period. It found that only 37 people had levels above 100 ng/ml. Only one person had true toxicity at 364 ng/ml.

     In one case study, a woman had a level of 476 ng/ml after taking a supplement that gave her 186,900 IU of vitamin D3 per day for two months. That is 47 times the generally recommended safe upper limit of 4,000 IU per day. This woman was admitted to the hospital after she experienced fatigue, forgetfulness, nausea, vomiting and slurred speech

     Although only extremely large doses can cause toxicity, even strong supporters of these supplements recommend an upper limit of 10,000 IU per day


    Elevated blood calcium levels

    Vitamin D helps your body absorb calcium from the food you eat.

    However, if vitamin D intake is excessive, blood calcium may reach levels that can cause unpleasant and potentially dangerous symptoms.


    Symptoms of high blood calcium levels (or hypercalcemia) include:

    o  digestive distress, such as vomiting, nausea, and stomach pain

    o  fatigue, dizziness and confusion

    o  excessive thirst

    o  and frequent urination


    Case studies

    The normal range of blood calcium is between 8.5 and 10.2 mg/dl.

     In one case study, an older man with dementia who received 50,000 IU of vitamin D daily for 6 months was repeatedly hospitalized with symptoms related to high calcium levels.

     In another, two men took improperly labeled vitamin D supplements, this lead to calcium levels of 13.2 and 15 mg/dl.

    It took a year for their levels to normalize after they stopped taking the supplements.

     

    Nausea, vomiting, and poor appetite

    Many side effects of too much vitamin D are related to excessive calcium in the blood.

     These include nausea, vomiting and poor appetite, however, these symptoms don't occur in everyone with elevated calcium levels.

     One study followed 10 people who had developed excessive calcium levels after they had taken high-dose vitamin D to correct a deficiency. Four of them experienced nausea and vomiting and three of them had a loss of appetite; so 7 out of 10 DID encounter some type of side effect

     Similar responses to vitamin D mega-doses have been reported in other studies. One woman experienced nausea and weight loss after taking a supplement that was found to contain 78 times more vitamin D than the dose stated on the label.

    Importantly, these symptoms occurred only in response to extremely high doses of vitamin D3, which led to calcium levels greater than 12 mg/dl.

     Get a blood test and only supplement when needed, you may not die from this, but you may be wasting good money on a supplement you don’t need.


    Stomach pain, constipation, or diarrhea

    Stomach pain, constipation and diarrhea are common digestive complaints that are often related to food intolerances or irritable bowel syndrome. However, they can also be a sign of elevated calcium levels caused by vitamin D toxicity.

     These symptoms may occur in those receiving high doses of vitamin D to correct a deficiency.

    As with other symptoms, response appears to be individualized even when vitamin D blood levels are similarly elevated.


    Case studies

     In one case study, a boy developed stomach pain and constipation after taking improperly labeled vitamin D supplements, whereas his brother experienced elevated blood levels without any other symptoms.

     In another case study, an 18-month-old child who was given 50,000 IU of vitamin D3 for 3 months experienced diarrhea, stomach pain and other symptoms.

    Thankfully, these symptoms subsided after the supplementation was curtailed.

     

    Bone Loss

    Because vitamin D plays an important role in calcium absorption and bone metabolism, getting enough is crucial for maintaining strong and healthy bones.

    However, too much vitamin D can be detrimental to bone health.

     Although many symptoms of excessive vitamin D are attributed to high blood calcium levels, some researchers suggest that mega-doses may also lead to low levels of vitamin K2 in the blood.

    This is important because one of vitamin K2's most important functions is to keep calcium in our bones and teeth and out of our sift tissues.

     To protect against bone loss, avoid taking excessive vitamin D supplements and take a vitamin K2 supplement too.

     You can also eat foods rich in vitamin K2, such as grass-fed dairy and meat, vegans may need to supplement.


    Kidney Injury

    Excessive vitamin D intake can also result in kidney injury.

    In one case study, a man was hospitalized for kidney failure, elevated blood calcium levels and other symptoms that occurred after he received vitamin D injections prescribed by his doctor.

    Indeed, most studies have reported moderate-to-severe kidney injury in people who develop vitamin D toxicity.

     In one study of 62 people who had received an excessively high-dose of vitamin D through injections; all 62 experienced kidney failure; whether they had healthy kidneys or an existing kidney disease.

     

    Even if you follow a healthy diet, you may require supplements to achieve optimal Vitamin D levels. However, it’s also possible to have too much of a good thing

    Make sure to avoid excessive doses of vitamin D. Generally, 4,000 IU or less per day is considered safe, but only as long as your blood values are being monitored.

    In addition, make sure you purchase supplements from reputable manufacturers to reduce the risk of accidental overdose due to inaccurate labeling.

     If you've been taking vitamin D supplements and are experiencing any of the symptoms I have mentioned, please consult a healthcare professional as soon as possible.

    The University of Minnesota Medical School and the United Health Group researchers found that metformin was associated with significantly reduced COVID-19 death risks in women in one of the world's largest observational studies of COVID-19 patients.


    Diabetes and COVID-19

    Severe COVID-19 outcomes for people with diabetes have been widely observed, including greater risk of intensive care unit admission, intubation for mechanical ventilation and death,

    These outcomes could possibly be related to less effective glycemic or blood sugar control in these patients.

    Metformin is an established, generic medication for managing blood sugar levels in patients with type 2 diabetes.

    It also reduces inflammation proteins like TNF-alpha that appear to make COVID-19 worse.

      

    The Study

    The team analyzed 6,256 individuals with type 2 diabetes or obesity who were hospitalized with COVID-19 and assessed whether or not the use of metformin was associated with decreased mortality

    The investigators identified 2,333 adults who had filled 90-day metformin prescriptions within 12 months of their COVID-19 diagnosis (with a mean age 73), and 3,923 non-users of Metformin (with a median age of 76)

    They then assessed the associations between home metformin use and risk of in-hospital death from COVID-19.

     

    The results

    They found an association that women with diabetes or obesity, who were hospitalized for COVID-19 disease and who had filled a 90-day metformin prescription before hospitalization, had a 21% to 24% reduced likelihood of mortality compared to similar women not taking the medication.

    There was no significant reduction in mortality among men.

     

    Carolyn Bramante, MD, MPH, the principal investigator and assistant professor in the Department of Medicine at the University of Minnesota Medical School said "Observational studies like this cannot be conclusive, but contribute to growing bodies of evidence. Seeing a bigger association with protection in women over men may point towards inflammation reduction as a key way that metformin reduces risk from COVID-19. However, more research is needed,"

    She also said:

    "A large database covering different geographic areas is rarely available. We were fortunate to have the opportunity to do this research alongside UnitedHealth Group."

    Deneen Vojta, MD, executive vice president of Enterprise Research and Development at UnitedHealth Group said, "While effective therapies to mitigate the harm of the SARS-CoV-2 virus are being developed, it is important that we also look to, and evaluate commonly used medications with good safety profiles for their potential to combat the virus,"

    New Drug Application

    Christopher Tignanelli, MD, assistant professor in the Department of Surgery at the University of Minnesota Medical School and Carolyn Bramante, together have submitted an investigational new drug application to the Food and Drug Administration for use of metformin for COVID-19 treatment and prevention.

     The FDA approved this application.

    The antioxidant quercetin is sometimes called the “Head or Master Flavonoid.” People have said it is one of the best natural antihistamines on the market. It can, allegedly, stabilize mast cells and reduce inflammation.

    A Mast cell serves the same general functions in the body and central nervous system; things such as effecting or regulating allergic responses, innate and adapted immunity, autoimmunity and inflammation.

    What is Quercetin?

    Quercetin is a plant flavanol from the flavonoid group of polyphenols.

    It’s one of the most abundant antioxidants in the diet and plays an important role in helping your body combat the free radical damage that has been linked to some chronic diseases.

    Antioxidants

    Antioxidants are compounds that can bind to and neutralize free radicals. Free radicals are unstable molecules that may cause cellular damage when their levels become too high

    Damage caused by free radicals has been linked to numerous chronic conditions, including cancer, heart disease and diabetes

     In addition, its antioxidant properties may help reduce:

    o  inflammation

    o  allergy symptoms

    o  blood pressure

    Food Sources

    Foods that commonly contain quercetin include:

    o  Onions

    o  Apples

    o  Grapes

    o  Berries

    o  Broccoli

    o  Citrus fruits

    o  Cherries

    o  Green tea, and for me thankfully coffee and red wine

    Inflammation

    Free radicals may do more than simply damage your cells.

    Research shows that high levels of free radicals may help activate genes that promote inflammation, ergo high levels of free radicals may lead to an increased inflammatory response.

    While a little inflammation is necessary to help your body heal and fight infections, persistent inflammation is linked to health problems, including certain cancers, as well as heart and kidney disease.

    Studies show that quercetin may help reduce inflammation.

    In test-tube studies, quercetin reduced markers of inflammation in human cells, including the molecules ‘tumor necrosis factor alpha (TNFα)’ and interleukin-6, both of these are a pro-inflammatory cytokines.

    Scientific Study

    An 8-week study of 50 women with rheumatoid arthritis observed that participants who took 500 mg of quercetin a day experienced a significant reduction in:

    o  Early morning stiffness

    o  Morning pain

    o  And after-activity pain

     They also had reduced markers of inflammation, such as TNFα, compared to those who received a placebo.

    While these findings are promising, more human research is needed to fully understand the compound’s potential anti-inflammatory properties.

    Allergies

    Quercetin’s potential anti-inflammatory properties may also provide allergy symptom relief.

    Test-tube and animal studies found that it may block enzymes involved in inflammation and suppress inflammation-promoting chemicals, such as histamine.

    For example, one study showed that taking quercetin supplements suppressed peanut-related anaphylactic reactions in mice.

    Still, it’s unclear whether the compound has the same effect on allergies in humans, so more research is needed before it can be recommended as an alternative treatment.

    Cancer

    Quercetin may have anti-cancer effects because it has certain antioxidant properties.

    In a review of test-tube and animal studies, quercetin was found to suppress cell growth and induce cell death in prostate cancer cells.

    Other test-tube and animal studies observed that the compound had similar effects in liver, lung, breast, bladder, blood, colon, ovarian, lymphoid and adrenal cancer cells.

     Though these findings are promising, many more human studies are needed before quercetin can be recommended as an alternative treatment for cancer.

    Brain Disorders

    Quercetin may lower your risk of chronic brain disorders. Research suggests that quercetin’s antioxidant properties may help protect against degenerative brain disorders, such as Alzheimer’s and dementia.

    In one study, mice with Alzheimer’s disease received quercetin injections every 2 days for 3 months. By the end of the study, the injections had reversed several markers of Alzheimer’s, and the mice performed much better during learning tests.

    In another study, a quercetin-rich diet reduced the markers of Alzheimer’s disease and improved brain function in mice at the early to middle stage of the condition. However, the diet had little to no effect on animals with middle to late stage Alzheimer’s.

    Coffee is a popular beverage that has been linked to a lower risk of Alzheimer’s disease. Again, research shows that quercetin, not caffeine, is the primary compound in coffee that’s responsible for its potential protective effects against this illness.

    Though these findings are promising, more research in humans is needed.

    Blood Pressure

     Quercetin may also reduce blood pressure, high blood pressure affects 1 in 3 American adults. It raises your risk of heart disease and is the leading cause of death in the United States.

     When mice with high blood pressure were given quercetin daily for 5 weeks, their systolic and diastolic blood pressure values decreased by an average of 18% and 23%, respectively.

     A review of 9 human studies in 580 people found that taking more than 500 mg of quercetin a day in supplement form, reduced systolic and diastolic blood pressure by an average of 5.8 and 2.6, respectively.

     Although these findings are promising, more human studies are needed to determine whether the compound could be an alternative therapy for treating high blood pressure

    Quercetin Supplements

    You can purchase quercetin as a dietary supplement, indeed DoNotAge.org now carry it in capsules form.

    Typical dosages range from 500 to 1,000 mg per day.

     Additionally, some research indicates that quercetin has a synergistic effect when quercetin is combined with other flavonoid supplements, such as resveratrol or Genistein.

    Side effects

    Quercetin is found naturally in many fruits and vegetables and so is safe to consume and a supplement, it appears to be generally safe with little to no side effects.

    In some instances, taking more than 1,000 mg of quercetin per day may cause mild symptoms like headaches, stomach aches and/or or tingling sensations.

    When consumed in food, quercetin is safe for pregnant and woman who are breastfeeding.

    However, studies on the safety of quercetin as a supplement are few, so you should avoid taking quercetin supplements if you’re pregnant or nursing.

     As with any supplement, consult your healthcare provider before taking quercetin, as it can interact with some medications, including antibiotics and blood pressure medications.

    The Sagol Center for Hyperbaric Medicine and Research at Shamir Medical Center, Tel Aviv University announced that according to a new study, for the first time in humans, two key biological hallmarks of aging, telomere length shortening, and the accumulation of senescent cells have been reversed.

    Hallmarks of Aging

    The biological deterioration of aging is cited as a major risk factor for cancer, cardiovascular diseases, diabetes, dementia and Alzheimer's.

    At the cellular level, two key hallmarks of the aging process are:

    The shortening of telomere length, of approximately 20-40 bases per year, which is associated with a variety of serious life-threatening illnesses, and

    The accumulation of senescent cells, the so-called "old malfunctioning cells," which inhibit cell proliferation. The accumulation of senescence contributes to many age-associated conditions and illnesses, while elimination of those cells can reverse them, as shown in previous animal studies.

    The Study Team

    The clinical trial was conducted by Prof. Efrati, MD, from the Faculty of Medicine and Sagol School of Neuroscience at Tel Aviv University, and Dr. Amir Hadanny, MD, Chief Medical Research Officer of The Sagol Center for Hyperbaric Medicine and Research and co-author of the study.

    It is the first study to evaluate whether HBOT can affect telomere length and senescence using their specific Hyperbaric Oxygen Therapy HBOT protocol.

    The Study

    The scientific included 35 healthy adults aged 64 and older who did not undergo any lifestyle, diet or medication adjustments.

    Each patient was placed in a hyperbaric chamber for 90 minutes a day, for five days a week, over three months while breathing 100% oxygen through a mask.

    The pressurized chamber allows more oxygen to be dissolved into the tissues and mimics a state of hypoxia, or oxygen shortage, which is known to have regenerating effects.

    Whole blood samples were collected prior to treatment, at the 30th and 60th session, and one to two weeks following the last HBOT session, to assess peripheral blood mono-nuclear cells telomere length and senescence.

    Statement

    In a statement Prof. Shai Efrati said, "After dedicating our research to exploring its impact on the areas of brain functionality and age-related cognitive decline, we have now uncovered for the first time in humans, Hyperbaric Oxygen Therapy's biological effects at the cellular level in healthy aging adults."

    Since telomere shortening is considered the 'Holy Grail' of the biology of aging, many pharmacological and environmental interventions are being extensively explored in the hopes of enabling telomere elongation.

    The significant improvement of telomere length shown during and after these unique Hyperbaric Oxygen Therapy protocols provides the scientific community with a new foundation of understanding that aging can, indeed, be targeted and reversed at the basic cellular-biological level.

    Results

    Results found that the telomere length of T helper, natural killer and B cells increased significantly, by over 20% following HBOT.

     The most significant change was in B cells, which increased at the 30th session, 60th session and post HBOT by 25.68%, 29.39% and 37.63% respectively.

    In addition, there was a significant decrease in the number of senescent T helpers by -37.30%, www.Eurekalert.com website summarized the results as a lengthening of up to 38% of the telomeres, as well as a decrease of up to 37% in the presence of senescent cells.

    Statement

    It’s a known fact that Dark chocolate is loaded with nutrients that can positively affect your health.

    Made from the seed of the cocoa tree, it is one of the best sources of antioxidants on the planet.

    Studies show that dark chocolate (not milk chocolate) can improve your health and lower the risk of heart disease.

    Nutrition

    Dark chocolate with a high cocoa content is actually very nutritious.

    A 100-gram bar of dark chocolate with 70–85% cocoa contains:

    o  11 grams of fiber

    o  67% of the RDI for iron

    o  58% of the RDI for magnesium

    o  89% of the RDI for copper

    o  98% of the RDI for manganese

    It also has plenty of potassium, phosphorus, zinc and selenium.

    Of course, 100 grams (3.5 ounces) is a fairly large amount and all these nutrients also come with 600 calories, for this reason, dark chocolate is best consumed in moderation.

     The fatty acid profile of cocoa and dark chocolate is also excellent. The fats are mostly saturated and monounsaturated, with small amounts of polyunsaturated fat.

     It also contains stimulants like caffeine, but is unlikely to keep you awake at night as the amount of caffeine is very small compared to coffee.

    Antioxidants

    Powerful Source of Antioxidants

    ORAC stands for “oxygen radical absorbance capacity.” It is a measure of the antioxidant activity of foods. This is where researchers set a group of free radicals (bad) against a sample of a food and see how well the antioxidants in the food can “disarm” the radicals.

     The biological relevance of ORAC values is questioned, because it’s measured in a test tube and may not have the same effect in the body. However, it is worth mentioning that raw, unprocessed cocoa beans are among the highest-scoring foods that have been tested.

    Dark chocolate is loaded with organic compounds that are biologically active and function as antioxidants. These include polyphenols and among others.

    One study showed that cocoa and dark chocolate had more antioxidant activity, polyphenols and flavanols than any other fruits tested, which included blueberries and acai berries.

    Blood Flow & Pressure

    May Improve Blood Flow and Lower Blood Pressure

    The flavanols in dark chocolate can stimulate the endothelium, the lining of arteries, to produce nitric oxide, one of the functions of nitric oxide is to send signals to our arteries to relax, which lowers the resistance to blood flow and therefore reduces blood pressure.

    Many controlled studies show that cocoa and dark chocolate can improve blood flow and lower blood pressure, though the effects are usually mild.

     HDL & LDL

    Consuming dark chocolate can improve several important risk factors for heart disease.

    In a controlled study, cocoa powder was found to significantly decrease oxidized LDL cholesterol in men. It also increased HDL and lowered total LDL for those with high cholesterol.

     Oxidized LDL means that the LDL had reacted with free radicals; this makes the LDL particle itself reactive and capable of damaging other tissues, such as the lining of the arteries in your heart.

     It makes sense that cocoa lowers oxidized LDL as it contains an abundance of powerful antioxidants that do make it into the bloodstream and protect lipo-proteins against oxidative damage.

    Dark chocolate can also reduce insulin resistance, which is another common risk factor for many diseases like heart disease and diabetes.

    Heart Disease

    May Reduce Heart Disease Risk

    In a study of 470 elderly men, cocoa was found to reduce the risk of death from heart disease by 50% over a 15-year period.

    Another study revealed that eating chocolate two or more times per week lowered the risk of having calcified plaque in the arteries by 32%.

     Another study showed that eating dark chocolate more than 5 times per week lowered the risk of heart disease by 57%

     Sun Damage

    May Protect Your Skin from the Sun.

    The bioactive compounds in dark chocolate may also be great for your skin. The flavanols can protect against sun damage, improve blood flow to the skin and increase skin density and hydration.

     The minimal erythemal dose (or MED) is the minimum amount of UVB rays required to cause redness in the skin 24 hours after exposure. In one study of 30 people, the MED more than doubled after consuming dark chocolate high in flavanols for 12 weeks.

     So, maybe consider loading up on dark chocolate prior to your beach vacation.

    Brain Function

    More good news, dark chocolate may also improve the function of your brain.

    One study of healthy volunteers showed that eating high-flavanol cocoa for five days improved blood flow to the brain.

     Cocoa may also significantly improve cognitive function in elderly people with mental impairment. It may improve verbal fluency and several risk factors for disease.

    Additionally, cocoa contains stimulant substances like caffeine, which may be a key reason why it can improve brain function in the short term.

    There is considerable evidence that cocoa can provide powerful health benefits, being especially protective against heart disease.

    Of course, this doesn’t mean you should go all out and consume lots of chocolate every day. It is still loaded with calories and easy to overeat.

     Also be aware that a lot of the chocolate on the market is not healthy, but good quality dark chocolate with 70% or higher cocoa content.

     Dark chocolates does contain some sugar, but the amounts are usually small and the darker the chocolate, the less sugar it will contain

    A study by the Boston University has shown that having sufficient levels of Vitamin D can prevent the conditions of Covid-19 patients’ from deteriorating and it can even reduce their need for oxygen.

    The researchers at the Boston University stated that only 9.7% of patients who were older than 40 years of age and who were vitamin D sufficient succumbed to the infection. Compared to 20% of people who had a circulating level of <30 nanograms per milliliter of 25-hydroxy-vitamin D; a measure of Vitamin D in the body.

    CRP or C reactive protein is commonly analyzed to check the inflammation levels caused by an infection. The higher the inflammation, usually the greater the infection

    A significant reduction in serum CRP, an inflammatory marker, suggests that vitamin D sufficiency also may help modulate the immune response, possibly by reducing risk of a cytokine storm in response to this viral infection.

    Boston University School of Medicine

    During the study, the Vitamin D levels of 235 hospitalized COVID-19 patients were measured.

    “This study provides direct evidence that vitamin D sufficiency can reduce the complications, including the cytokine storm and ultimately death from COVID-19.”

    A cytokine is a small protein important in the cell signaling process.

    A cytokine storm is where too many proteins are released into the blood too quickly, which can be deadly.

    Vitamin D and the immune response

    Patients who were Vitamin D sufficient had significantly lower blood levels of the inflammatory marker CRP and had a higher total blood lymphocyte count; this suggests that Vitamin D sufficiency had improved their immune function.

    The researchers concluded that the beneficial effect on the immune system may also reduce the risk of acquiring this potentially life-threatening viral infection in the first place.

    Only 32.8% of participants in the study had sufficient Vitamin D levels.

    The severity of clinical outcomes from COVID-19 and mortality were reduced in patients who were vitamin D sufficient.

    Clinical features were also significantly different in patients who were Vitamin D sufficient, they had a lower risk of becoming unconscious and becoming hypoxic.

    Vitamin D is really not a vitamin. Vitamins are special nutrients that the body needs but cannot make, so they must be obtained from what we eat or by supplements. Because our bodies can make Vitamin D in our skin when it is exposed to sunlight, Vitamin D is considered a hormone.

     

    Vitamin D2 and Vitamin D3

    Vitamin D is actually available in two forms: vitamin D2 and vitamin D3; the main difference really is just the source.

    ·     D2 is manufactured from plants and fungi (and often the type that’s included in fortified milk, bread, and cereal)

    ·     D3 is created from animal products and is the closest to that which is naturally made by the body through sunlight exposure.

    Because of this familiarity, most doctors and nutritionists recommending supplementing with vitamin D3.


     The Benefits of Vitamin D3

    Vitamin D3 can have an impact on as many as 2,000 different genes within the body, and has a direct role on far more aspects of health than many might realize.

    ·     Vitamin D receptors are found in nearly every cell, and as soon as Vitamin D binds to a receptor, it turns genes on or off, prompting changes at the cellular level

    ·     Studies completed over the last two decades have proven that this process turns off cancer-causing genes, turns on immune-protective genes, and even tells cells which vitamins and minerals to absorb.

    This all has a great effect on many important body processes, let’s look at a few:

    Food Sources of Vitamin D

    Very few foods actually contain vitamin D naturally.

    Because of this, some foods are fortified, which means vitamin D is added during the production process.

    Foods that do contain vitamin D include:

    ·      salmon

    ·      sardines

    ·      egg yolk

    ·      shrimp

    ·      milk (fortified)

    ·      yogurt (fortified)

    It can be hard to get enough vitamin D each day through sun exposure and food alone, so taking vitamin D supplements can help

     

    Bone Health

    Correct Vitamin D3 levels result in strong bones and teeth, because it helps regulate and control the body’s ability to absorb phosphorus and calcium.

    ·     Phosphorus and calcium provide density and strength to both the skeletal system and teeth.

    ·     Vitamin D deficiency can lead to a loss of bone density, which can contribute to osteoporosis and fractures (broken bones).

    ·     Severe vitamin D deficiency can also lead to other diseases. In children, it can cause rickets.

    Rickets is a rare disease that causes the bones to become soft and bend.

     

    Autoimmune system:

    Vitamin D modulates our innate and our adaptive immune responses.

    ·     Deficiency in vitamin D is associated with increased autoimmunity as well as an increased susceptibility to infection.

    ·     One study showed evidence that Vitamin D Supplementation Could Reduce Risk of Influenza and COVID-19 Infections and Deaths

    ·     The link between Vitamin D deficiency and autoimmune disease is well documented, which means if you are deficient in Vitamin D you are more likely to contract an autoimmune disease

     

    Cancer Prevention

    ·     Some evidence supports the idea that increased vitamin D3 can help slow the progression of prostate tumors, while other reports show promise that combining the nutrient with more fiber can reduce the risk of developing polyps that could lead to colon cancer

    ·     Another helpful combo is vitamin D and calcium: A four-year trial found that when post-menopausal women supplemented with both, their likelihood of developing cancer dropped by 60%

    ·     Vitamin D has also been frequently studied for its apparent relationship to breast cancer: In a study of 166 women undergoing treatment, nearly 70 percent had low levels of vitamin D, which led many scientists to theorize that there could be a link.

     

    Weight loss

    Consider adding vitamin D supplements to your diet if you’re trying to lose weight.

    ·     In one study, people taking a daily calcium and vitamin D supplement were able to lose more weight than subjects taking a placebo supplement. The scientists said the extra calcium and vitamin D had an appetite-suppressing effect.

     

    How much do you need?

    There has been some controversy over the amount of vitamin D needed for healthy functioning. Recent research indicates that you need more vitamin D than was once thought.

    ·     Normal blood serum levels range from 20, but some organisations say 50, to 100 micrograms per deciliter. But, depending on your blood level, you may need more vitamin D.

    The Institute of Food and Agricultural Sciences recommended daily IU intake for vitamin D are:

    ·     children and teens: 600 IU

    ·     adults up to age 70: 600 IU

    ·     adults over age 70: 800 IU

    ·     pregnant or breastfeeding women: 600 IU

     

    Personally I had a blood test to see what my levels were and then started to supplement with Vitamin D3 and K2. I take 5,000 IU of Vitamin D3 a day.

    I recommend that before you start to supplement you have a blood test and then consult a doctor to see what level of supplementation would be favorable.

    You may not need to take a supplement if your levels are high enough.


    Low Magnesium

    A review published in The Journal of the American Osteopathic Association found Vitamin D can't be metabolized without sufficient magnesium levels, meaning Vitamin D remains stored and inactive for as many as 50 percent of Americans.

    ·     Patients with optimum magnesium levels require less Vitamin D supplementation to achieve sufficient Vitamin D levels.

    ·     Deficiency in either of these nutrients is reported to be associated with various disorders, including skeletal deformities, cardiovascular diseases and metabolic syndrome

    ·     Research shows that magnesium consumption from natural foods has decreased in the past few decades, owing to industrialized agriculture and changes in dietary habits

    ·     The Magnesium status is low in populations who consume processed foods that are high in refined grains, bad fats, phosphate and sugar

    Silver Bullet

    As you may know I started NMN experiment to see how NMN worked, but within weeks of getting into it I realized that taking NMN as a daily supplement was not going to be a silver bullet, there are many other things that are key to extending our Health-span

    So, as well as NMN supplementation I will also continue to make videos, like this, on other subjects that track my other longevity ‘hacks’ if you like,

    Most people have never heard of vitamin K2.

    Vitamin K2 is rare in the Western diet and hasn't received much mainstream attention; however this powerful nutrient plays an essential role in many aspects of our health.


    What Is Vitamin K?

    Vitamin K was discovered in 1929 as an essential nutrient for blood clotting.

    ·     The initial discovery was reported in a German scientific journal, where it was called “Koagulations-vitamin” — which is where the "K" comes from

    ·     It was also discovered by the dentist Weston Price, who travelled the world in the early 20th Century studying the relationship between diet and disease in different populations

    ·     He found that the non-industrial diets were high in some hitherto un-identified nutrient, which seemed to provide protection against tooth decay and chronic diseases

    Vitamin K1 & K2

    There are two main forms of vitamin K:

    ·      Vitamin K1, which is found in plant foods like leafy greens

    ·      Vitamin K2, which is found in some animal and fermented foods

    Vitamin K2 can be further divided into several different subtypes, the most important ones being MK-4 and MK-7


    How Do Vitamins K1 and K2 Work?

    Vitamin K activates proteins that play an important role in blood clotting, calcium metabolism and our heart health

    ·     One of its most important functions is to regulate calcium deposition. In other words, it promotes the calcification of bones and prevents the calcification of our soft tissues such as blood vessels and kidneys

    ·     Some scientists have suggested that the roles of vitamins K1 and K2 are very different, and many feel that they should be classified as two separate nutrients, this idea is supported by an animal study showing that vitamin K2 (MK-4) reduced blood vessel calcification whereas vitamin K1 did not.

    ·     Controlled studies in people also observe that vitamin K2 supplements generally improve bone and heart health, while vitamin K1 shows no significant benefits

    However, more human studies are needed before the functional differences between vitamins K1 and K2 can be fully understood

     Heart Disease

    ·     In one study spanning 10 years, people with the highest intake of vitamin K2 were 52% less likely to develop artery calcification and had a 57% lower risk of dying from heart disease

    ·     Another study in 16,057 women found that participants with the highest intake of vitamin K2 had a much lower risk of heart disease - for every 10 micrograms of K2 they consumed per day, the risk of heart disease was reduced by 9%

    It is generally agreed that more long-term controlled trials on vitamin K2 and heart disease are needed.

    Bone Health

    Vitamin K2 may Help Improve Bone Health and Lower Your Risk of Osteoporosis

    ·     Osteoporosis, which translates as “porous bones”, is a common problem in many Western countries. It is prevalent among older women and strongly raises the risk of fractures.

    As I mentioned before vitamin K2 plays a central role in the metabolism of calcium; calcium being the most abundant mineral found in our bones and teeth

    ·     Vitamin K2 activates the calcium-binding actions of two proteins; the matrix GLA protein and osteocalcin, which help to build and maintain bone cells

    ·     A 3-year study in 244 post-menopausal women found that those taking vitamin K2 supplements had a much slower decrease in age-related bone mineral density

    Japanese studies

    ·     Long-term studies in Japanese women have observed similar benefits, although very high doses were used in these cases. Out of 13 studies, only one failed to show significant improvement.

    ·     Seven of these trials, which took fractures into consideration, found that vitamin K2 reduced spinal fractures by 60%, hip fractures by 77% and all non-spinal fractures by 81%

    ·     In line with these findings, vitamin K supplements are officially recommended for preventing and treating osteoporosis in Japan

    Dental health

    Researchers have speculated that vitamin K2 may also affect dental health; however no human studies to date have tested this directly.

    ·     Based on animal studies and the role vitamin K2 plays in bone metabolism, it’s reasonable to assume that this nutrient impacts dental health as well

    ·     One of the main regulating proteins in dental health is osteocalcin, this is the same protein that is critical to bone metabolism and is activated by vitamin K2

    ·     Osteocalcin triggers a mechanism that stimulates the growth of new dentin, which is the calcified tissue underneath the enamel of your teeth

    Cancer

    Unfortunately, as we all know cancer is a common cause of death worldwide.

    ·     Even though modern medicine has found many ways to treat it, new cancer cases are still on the rise. Therefore, finding effective prevention strategies is vital

    ·     Two clinical studies suggest that vitamin K2 reduces recurrence of liver cancer and increases survival times

    ·     Additionally, an observational study in 11,000 men found that a high vitamin K2 intake was linked to a 63% lower risk of advanced prostate cancer, whereas vitamin K1 had no effect

    That said, it is widely acknowledged that more high-quality studies are needed before any definitive claims can be made.

    How to get it

    Several widely available foods are rich sources of vitamin K1, but vitamin K2 is less common.

    ·     Your body can partly convert vitamin K1 into K2. This is useful, as the amount of vitamin K1 in a typical diet is ten times that of vitamin K2.

    However, current evidence indicates that the conversion process is inefficient. And as a result, you may benefit much more from supplementing with vitamin K2

    ·     Vitamin K2 is also produced by gut bacteria in your large intestine. Some evidence suggests that broad-spectrum antibiotics can contribute to K2 deficiency

    ·     Vitamin K2 is mainly found in fermented foods, which unfortunately most people don't eat

    ·     Rich animal sources include high-fat dairy products from grass-fed cows, egg yolks, as well as liver and other organ meats, which again most people don't eat

    ·     The benefits of supplementing with K2 may be enhanced even further when combined with a vitamin D3 supplement, as these two vitamins have strong synergistic effects

    What is Fisetin?

    ·     Fisetin is a flavonol, a yellow plant pigment that belongs to the flavonoid group of polyphenols. It gives color to many different fruits and vegetables.

    ·     Scientists are currently exploring its ability to slow the aging process and extend lifespan, so basically its senolytic effects, which means can Fisetin selectively induce the death of senescent cells and improve health in humans

    What’s more, fisetin has powerful anti-inflammatory, antioxidant, and immune-supporting properties

    Despite promising animal and cell-based findings, research is still in its early stages. Only three clinical trials to date with humans

    Food Sources

    Many different fruits and vegetables contain fisetin.

    Food sources with the highest concentration of fisetin include:

    ·     Strawberries (160)

    ·     Apples (27)

    ·     Persimmons (11)

    ·     Lotus root (6)

    ·     Onions (5)

    ·     Grapes (4)

    ·     Kiwi (2)

    (the number in brackets relates to the micrograms value per gram of freeze-dried food)

    Blocks the NF-κB Pathway

    ·     The NF-κB pathway is a switch that tells genes to produce inflammatory compounds

    ·     An overactive NF-κB response is linked to allergies, autoimmune diseases and some cancers. Plus, fisetin blocks inflammatory enzymes that degrade fatty acids.

    ·     It also reduces the activity of a group of enzymes that cancers require to spread and invade other tissues

     Blocks the mTOR pathway

    ·     mTOR’s demands for energy and growth can throw your cells into a frenzy: metabolic waste builds up – and there’s no time to clean it up.

    ·     An overactive mTOR response is associated with cancer, diabetes, obesity, and neurodegenerative diseases. By blocking this pathway, fisetin helps remove waste and enhance cellular metabolism

    ·     And here’s the most intriguing part: blocking mTOR increases longevity. In fact, blocking mTOR is the only intervention that increased lifespan in all organisms studied to-date, this is because once mTOR is blocked, autophagy is activated: a process of recycling damaged cellular components

    Please bear in mind that the vast majority of fisetin health effects haven’t yet been confirmed in clinical trials.

    Anti-Ageing

     The following only relate to animal and cell-based studies; they should help with future studies, but have not been proven in humans yet.

    ·     Aging is characterized by the buildup of senescent cells – cells that stop dividing, become damaged, and start releasing inflammatory molecules

    ·     As we accumulate more and more of these senescent cells, the body starts being affected. These cells start damaging healthy tissue. They contribute to many age-related diseases – from osteoporosis and cancer to heart and brain diseases, Removing senescent cells calms inflammation, improves physical function, and increases lifespan in animals

    ·     Certain plant compounds are able to destroy senescent cells without harming healthy cells. In a 2018 cell-based study of 10 such compounds, fisetin was the most effective one. In old mice, fisetin cleared senescent cells and increased their lifespan by over 10%

    ·     Fisetin also extended the lifespan of fruit flies by more than 20% and yeast by more than 50%, as a result of this a clinical trial is underway to see if fisetin is effective for reducing inflammation and improving frailty and bone health in elderly people

     Diabetes and Its Complications

    In multiple animal studies, fisetin restored blood sugar levels of diabetic rats and mice to those of healthy animals. It improved their ability to control blood sugar levels by:

    • Increasing insulin levels
    • Increasing enzymes that turn sugar into energy
    • Removing sugar from the blood to store as glycogen in the liver
    • Reducing the liver’s ability to make sugar from lactate and amino acids

    ·     Fisetin slowed the progression of cataracts and protected the kidneys of diabetic mice by blocking inflammation and oxidative stress. It also protected the liver of diabetic rats from high blood sugar levels by increasing antioxidant levels

    ·     Another complication of diabetes is the hardening of the arteries and heart disease. In a cell study, fisetin prevented high sugar levels from causing inflammation in blood vessels

     Anticancer Potential

    The findings discussed below stem from preliminary clinical research and animal studies. Fisetin isn’t approved for cancer prevention or treatment.

    ·     Inflammation is linked to colon cancer growth, as well as it’s spread and resistance to chemotherapy. In a clinical study of 37 colon cancer patients undergoing chemotherapy, fisetin (100 mg/day for seven weeks) reduced markers of inflammation.

    ·     In rats, fisetin reduced oxidative stress and the growth of liver cancer caused by fungal toxins

    ·     In mice, it prevented the growth of lung cancer and boosted low antioxidant levels caused by a toxin in tobacco smoke. It reduced lung tumor growth by 67% in mice and by 92% when combined with a chemotherapy drug. It also prevented the growth of new blood vessels supplying nutrients to the cancer

    ·     Another study found that fisetin reduced tumor growth by 66% in mice with melanoma

    ·     Certain types of prostate cancers are fueled by androgens such as testosterone, fisetin slowed the growth of prostate tumors in mice by blocking the receptors for testosterone on cancer cells

    ·     Fisetin also protected against kidney damage from chemotherapy in rats by reducing inflammation and boosting antioxidant levels

    Cognition

    ·     Older rats given fisetin experienced memory and learning improvements.

    ·     Cell studies revealed that fisetin activates pathways in the brain involved in storing memories

    In another study, fisetin prevented memory loss in mice exposed to toxins

    ·    Fisetin can easily cross the blood-brain barrier in mice. This is important as there is an ongoing debate about whether compounds like fisetin can reach high enough levels in the brain to improve its function

    Neurodegenerative Diseases

    When over activated Immune cells in the brain cause Neurodegenerative diseases such as Alzheimer’s and Huntington’s.

    ·     This causes inflammation and damage to healthy brain cells. In cells studies, fisetin boosted brain antioxidant levels and prevented these immune cells from releasing inflammatory compounds in response to bacterial toxins

    ·     Alzheimer’s disease involves the buildup of amyloid plaques and tau proteins in the brain. Fisetin reduced levels of tau proteins in brain cells by activating a process that removes these harmful proteins known as autophagy

    ·     In mice with amyloid plaques, fisetin improved memory, reduced inflammation, and prevented the loss of brain cell function. In mice with Alzheimer’s, it reduced amyloid plaque buildup and loss of brain cells.

    ·     Huntington’s disease is a genetic movement disorder in which brain cells get destroyed. In mice with Huntington’s, fisetin was able to improve declining physical function and increase lifespan

    Cholesterol

    Fisetin reduced high total and LDL cholesterol and triglycerides in rats fed a high-fat diet.

    ·     In diabetic rats, it doubled HDL levels and cut LDL cholesterol levels in half. A cell study hinted that fisetin reduces cholesterol by causing more of it to be released in the bile

    ·     Cell studies show that fisetin prevents immune cells called macrophages from oxidizing and ingesting LDL cholesterol. When macrophages ingest oxidized LDL, they create fatty plaques that harden the arteries and cause heart disease

    Bone Loss

    ·     Estrogens keep bones healthy. After menopause, low estrogen levels put women at risk of osteoporosis. The rise inflammation due to aging also weakens the bones

    ·     Fisetin improved bone density and prevented bone loss in mice with low estrogen levels and inflammation. In cells, it worked by reducing the activity of bone-degrading cells (osteoclasts)

     So Gentlemen, it’s unlikely you will have heard of this supplement, although the ladies in your life most probably will have and are probably using it too.

    So, in the beauty industry Hyaluronic Acid or (HA) is used in a bid to maintain moisture levels in the skin; which I turn leads to smooth, flawless, hydrated and younger looking skin, well that’s the claim.

    I won’t get into the hugely inflated cost of HA products and the minuscule amount of HA that each product contains and that it doesn’t stay on your skin long enough to make a difference, maybe I’ll leave that for a future video?

    What is Hyaluronic Acid?

    ·     Hyaluronic acid, also known as hyaluronan, is a clear, gooey substance that is naturally produced by your body

    ·     The largest amounts of it can be found in your skin, connective tissue (such as bone, cartilage, fat etc.) and in your eyes

    ·     To keep the molecules in your skin, connective tissue and eyes well lubricated and moist its main function is to retain water,

    ·     In fact 1 gram of hyaluronic acid can retain up to 2 gallons (7.5 litres) of water, so it is a very powerful humectant

    Applications

    Hyaluronic acid or HA has a variety of uses, all of which we will cover shortly and it can be administered in a number of ways:

    ·    As a topical serums and in eye drops

    ·    As an injection

    ·    And a supplement

    Dr. Vera, First I want to talk quickly about Dr. Vera Gorbunova and her work in the antiaging field

    ·     She is a Professor of Biology at the University of Rochester, she is also the co-director of the Rochester Aging Research Center and she also sits on the DoNotAge.org advisory board.

    Her research is focused on understanding the mechanisms of longevity and genome stability, and on the studies of exceptionally long-lived mammals

    ·     She has spoken at TEDx in Cannes and at the Undoing Aging conference in Berlin; links in the description below to these talks, both extremely interesting and well worth a watch

    ·     The naked mole rat lives on average for 30 years; the longest lived rodent by far. The average mouse lives for only 4 years.

    Dr Vera Gorbunova concludes that this may be because of their abundance of High Molecular Weight Hyaluronic Acid or HMW-HA

    ·     Naked mole rats very, very rarely get cancer and Vera and her team found that removing the HMW-HA from her naked mole rats makes them more prone to cancer

    So, it appears that the more HMW-HA, the less likely your cancer risk.

    Please note this study was done in rodents and that human trials are not yet concluded

    Hyaluronic Acid Promotes Healthier and More Supple Skin

    ·     Hyaluronic acid supplements can help your skin look and feel suppler. Roughly half of the hyaluronic acid in your body is present in your skin, where it binds to water to help retain moisture.

    ·     However, the natural aging process and exposure to things such as ultraviolet radiation from the sun, tobacco smoke and pollution can decrease the amount in your skin. Taking hyaluronic acid supplements may prevent this decline by raising the depleted levels

    ·     Doses of 120–240 mg per day for at least one month have been shown to significantly increase skin moisture and reduce dry skin in adults

    ·     Hydrated skin also reduces the appearance of wrinkles, which may explain why several studies show that supplementing with it can make skin appear smoother

     Study in Japan:

    ·     Sixty Japanese male and female subjects aged 22-59 years who presented with crow's feet were randomly assigned 120 mg/day or a placebo

    ·     The subjects were administered HA at a rate of 120 mg/day or a placebo for 12 weeks. The skin wrinkles were evaluated by image analysis of skin wrinkle replicas, and their skin condition was evaluated using a questionnaire

    ·     Results: During the study period, the HA groups showed better level of the whole sulcus volume ratio, wrinkle area ratio, and wrinkle volume ratio than the placebo group

    After 8 weeks of ingestion, the HA group showed significantly diminished wrinkles compared with the placebo group.

    After 12 weeks skin luster and suppleness significantly improved compared with their baseline

    ·     Conclusion: The results suggest that the HA supplementation inhibits skin wrinkles and improves skin condition

     Hyaluronic acid Can Speed Wound Healing

    Hyaluronic acid also plays a key role in wound healing.

    ·     It’s naturally present in the skin, but its concentrations increase when there is damage in need of repair

    ·     Hyaluronic acid helps wounds heal faster by regulating inflammation levels and signaling the body to build more blood vessels in the damaged area). Applying it to skin wounds has been shown to reduce the size of wounds and decrease pain faster than a placebo or no treatment at all

    ·     Hyaluronic acid also has antibacterial properties, so it can help reduce the risk of infection when applied directly to open wounds

    ·     What’s more, it’s also effective at combating gum disease, speeding up healing after tooth surgery and eliminating ulcers when used topically in the mouth

     Hyaluronic Acid can Relieve Joint Pain

    ·     Hyaluronic acid is also found in our joints; here it keeps the space between your bones well lubricated, when the joints are lubricated the bones are less likely to grind against each other and cause discomfort and pain

    ·     Hyaluronic acid supplements are very helpful for people suffering from osteoarthritis, a type of degenerative joint disease caused by wear and tear on the joints over time

    ·     Taking 80–200 mg daily for at least two months has been shown to significantly reduce knee pain in people with osteoarthritis, especially those between the ages of 40 and 70

    Hyaluronic Acid may Soothe Acid Reflux Symptoms

    ·     New research shows hyaluronic acid supplements may help reduce the symptoms of acid reflux.

    When acid reflux occurs, the contents of the stomach are regurgitated up into the throat, causing pain and damage to the lining of the esophagus. Hyaluronic acid may help soothe the damaged lining of the esophagus and speed up the recovery process.

    ·     One human study found that taking a hyaluronic acid and a kondrowatin sulfate supplement along with an acid-reducing medication decreased reflux symptoms 60% more than taking an acid-reducing medication alone.

    A second study showed that the same type of supplement was five times more effective at reducing acid reflux symptoms than a placebo

    Research in this area is still relatively new, and more studies are needed to replicate these results. Nevertheless, these outcomes are certainly promising

    Hyaluronic Acid Relieves Dry Eye and Discomfort

    ·     Approximately 1 out of 7 older adults suffer from the symptoms of dry eye because of reduced tear production or tears evaporating too quickly; hyaluronic acid is excellent at retaining moisture, it’s often used to treat dry eye

    ·     Eye drops containing 0.2 to 0.4% hyaluronic acid have been shown to reduce dry eye symptoms and improve overall eye health

    ·     Contact lenses that contain slow-release hyaluronic acid are also being developed as a possible treatment for dry eye.

    In addition, hyaluronic acid eye drops are frequently used during eye surgery to reduce inflammation and speed wound healing

    To date, no studies have examined the effects of hyaluronic acid supplements on dry eye, but it may be a future area of research

    Preserve Bone Strength

    New animal research has begun to investigate the effects of hyaluronic acid supplements on bone health.

    ·     Two studies have found that hyaluronic acid supplements can help slow the rate of bone loss in rats with osteopenia, the initial stage of bone loss that precedes osteoporosis

    ·     Test-tube studies have also shown that high doses of hyaluronic acid can increase the activity of osteoblasts, the cells that are responsible for building new bone tissue

    While its effects on human bone health have not yet been studied, early animal and test-tube studies are extremely promising

    Could Prevent Bladder Pain

    ·     Approximately 3 to 6% of women suffer from a condition called inter-stish-al cy-sti-tis, or painful bladder syndrome, this disorder causes abdominal pain and tenderness, along with a strong and frequent urge to urinate

    ·     While the causes of this condition are unknown, hyaluronic acid has been found to help relieve the pain and urinary frequency associated with this condition when inserted directly into the bladder through a catheter

    ·     It's unclear why hyaluronic acid helps relieve these symptoms, but researchers hypothesize that it helps repair damage to bladder tissue, making it less sensitive to pain

    Studies have not yet determined whether oral hyaluronic acid supplements can increase amounts of it in the bladder enough to have the same effects

    Possible Side Effects and Precautions

    Hyaluronic acid is generally very safe to use, with very few reported side effects

    ·     Since the body naturally produces it, allergic reactions are very rare, one study in 60 people with osteoarthritis who took 200 mg daily for one year reported no negative side effects

    ·     However, its effects during pregnancy or breastfeeding have not been thoroughly studied, so these groups should be cautious and avoid supplementing with it

     

    ·     There is also some evidence that cancer cells are sensitive to hyaluronic acid and taking supplements could make them grow faster, for this reason, it is generally advised that people with cancer or who have a family a history of cancer avoid supplementing with it

    ·     Hyaluronic acid injections into the skin or joints have a higher risk of side effects. However, negative reactions are mostly associated with the injection procedure, rather than hyaluronic acid itself

    Things I would and would not have changed:

    ·    See a doctor or health practitioner, I know that this will not be easy, a lots don’t know about LCHF let alone NMN, and still say things like LDL is your bad cholesterol, with no idea what VLDL is at all, so seek out one that is open to what you are doing

    ·    Would have gone from 0 to 1 gram quicker? Yes, but I only know this now because I did it slowly, there is no official dosage protocol, so we are all still guessing; Hindsight is a great thing and it may be different for you, so start small and work up at a pace you and more importantly your body are happy with

    ·    Blood tests as a baseline, get as many markers checked as you can afford (you know, this is not a bad thing to do anyway), I waited until I was increasing my dose to 1 gram a day; if I were starting again I would do it before day 1

    ·    Have a reliable way to check your biometric markers, if you can, buy a quality scale, my first gym almost doubling their prices overnight and the virus shutting down my second gym certainly wasn’t something I, or anyone, could have envisaged at the start of the experiment

    ·    If you can afford it, have a DEXA scan, possibly the most effective way to measure your bio-metric markers, although be aware you will be exposed to very minor levels of radiation during the procedure

    ·    Find out your biological age, use an Epi-genetic/DNA marker test, blood or saliva. Biometric scales use BMI as part of the formula to estimate the biological age, as you may know I do not hold any store in that antiquated formula

    ·    I would have taken TMG with NMN from the start, although not proven, methylation or depleting our methyl groups is something that we need to avoid

    ·    DNA now sell TMG, £15 (around $19) for 30 grams, again use my discount code ‘MYNMN’ to get 10% off

    ·    I would have added equal amounts of Resveratrol as I upped my dose of NMN, maybe 1 month after I was happy with no NMN side effects, something like this:

    22 April: 125 mg NMN*

    22 May: add 125mg of Resveratrol

    22 June: up to 250mg NMN*

    22 July: up to 250mg of Resveratrol, and so on.

    So, essentially stagger, the increase in dosage to see if there were any side effects.

    The asterisk denotes that the NMN would be taken with at least the same dose of TMG

    •     A little more research may have made the journey a little less fraught; but then you don’t know what you don’t know

    •     TMG, it wasn’t until I had been taking NMN for a few months that people, far more in the know than I, alerted me to methylation and the need to take TMG

    •     Resveratrol v Trans-resveratrol, again it wasn’t until someone mentioned it in the comments section and then David Sinclair confirmed (during an interview) that it must be Trans and not cis-resveratrol. I took cis-resveratrol for a few months before I knew this (this will have affected my results, to what extent I will never know)

    •     Berberine dose, Purely by luck I started to take one gram of Berberine in lieu of 1 gram of Metformin (as David Sinclair does), before I found out, again from a subscriber that 1 gram of Berberine is roughly equal to 1 gram of Metformin; Info from a Valter Longo video.

    •     Supplier, To date I have now NMN from 4 different suppliers, the latest be DoNotAge.org – I think that some diligent research on what suppliers to choose, so you don’t run out, maybe have 2 that you trust, that:

    •     Do show 3rd party test certificates (not will send on request)

    •     And can ship to your country (this was a big one for me)

    ·    Think about recording, as I did, things that you think may be affected by NMN, actually record them - i.e. sleep, exercise, hunger size etc. Also record things such as headaches, upset stomach etc. also record if they didn’t happen; remember no data is data

    ·    Don't start this with another big change; you may use taking NMN as part of a big life change i.e.

    o Take NMN and go carnivore or Vegan

    o Take NMN and starting to exercise regularly

    o Take NMN and giving up sugar

    ·    I am glad I was happy with my diet and exercise regime; big changes may have altered my Stat’s i.e. doing extra exercise, changing fasting regime etc. would have shown non-NMN triggered changes

    ·    Be aware of potential side effects, sleep issues, upset stomach, extra energy (don’t take at night) etc. Be aware that these things may happen

    Pharma grade or Food Grade

    ·     No Grades, there is no food grade, pharmaceutical grade of veterinary grade, there is just NMN, these grade labels are all unofficial and do not meet any official standard

    ·     CAS Registry Number, NMN has a CAS number, go to this website and enter in this number and it will show you all the information about NMN. I was told by a reliable source that, if there were more than 1 type of NMN, it would have a separate number.

    ·     Purity, the % of purity is the percentage of the desired material which is actually in the sample. It is not a standard, there is no standard that allows a company to call it’s NMN Pure, ultra pure, mega pure etc. these are labels made up by supplement companies, mainly to entice you, also to mislead (but within the law).

    The decision is up to you, are you happy with 95% purity or must you have 99%+ - Pharma grade, food grade, pure, super pure, ultra-pure are just labels, ignore them, just look at the percentage number, what is the difference between 99.5% and 95.5%, will the extra 4% actually make a difference at the molecular level, who knows? I’m sure the NMN companies, if asked, will certainly tell you ‘there is strong evidence to say it could or it may, it would be safer to take as pure a product as is possible; they don’t know, they don’t care, they just want your orders, they just want your money. Be sure you know what you are paying for, the purity level and not the unofficial label.

    ·     Cost, so basically, it is all down to cost, what can you afford? What purity do you think will have an effect?
